摘 要: | 甘蓝型油菜(AACC)与萝卜(RR)杂交以导入抗性基因,应用“胚胎挽救”技术在本研究中产生了133株F1代杂种株与油菜栽培品种回交的第1代杂种株,145株回交第2代杂种株.细胞学和同工酶(GPI,POX)酶谱分析表明,回交后代中萝卜的抗性R基因通过染色体工程可望导入到甘蓝型油菜栽培品种中.

The development and isozyme analysis of hybrids between Brassica napus and Raphanus sativus |

Abstract: | Since sexual hybridizations in the Cruciferae family between Brassica napus and Raphanus sativus is possible, it is feasible to transfer nematode resistant gene(s) from R. sativus into B. napus. In the present research, intergeneric sexual hybridization was applied to aid the transfer and the application of in vitro ovule culture confirmed that this embryo rescue technique was a successful tool to obtain hybrids from crosses involving B. napus and R. sativus. By using the embryo rescue technique 133 BC1 individuals derived from the cross of bridging hybrid allohexaploids (AACCRR 2n=56) × B.napus and 145 BC2 hybrids derived from the cross 〔allohexaploids(AACCRR) × B. napus〕× B. napus were obtained in different cross combinations. Cytological studies of BC1 individuals, which included both AACC and RR genome, indicated that the somatic chromosome numbers of BC1 individuals ranged from 42 to 47. The chromosome numbers of BC2 individuals ranged from 37 to 47. The BC1 and BC2 plants showed a large variation for both male and female fertility. The results of POX and GPI isozyme analysis suggested that the possibility to transfer important traits from Oilradish (Raphanus sativus L. ssp. oleiferus) to B. napus could be increased through following backcrosses. |
