摘    要:为探讨影响组培桑苗中1-脱氧野尻霉素(1-DNJ)合成的因素,采用反相高效液相色谱紫外检测法,调查了培养基成分对组培桑苗叶片中1-DNJ含量的影响。结果表明:采用MS培养基,适当提高果糖和无机盐浓度,降低磷酸盐浓度等,有利于桑苗叶片中1-DNJ的合成和积累;培养基中的Fe2+浓度对桑苗叶片中1-DNJ的含量有显著影响,无Fe2+时不能合成1-DNJ,Fe2+浓度在0.05~0.20 mmol/L范围内,1-DNJ的含量随着Fe2+浓度增加有所降低;培养基中的烟酸、维生素B6对于桑苗叶片中1-DNJ的合成具有抑制作用,而Mg2+、肌醇、维生素B1和甘氨酸等对1-DNJ合成的影响较为复杂,未见明显的规律性变化。

Abstract:To explore factors on the synthesis of 1-deoxynojirimycin in tissue culture mulberry seedling,we investigated the effect of medium compositions on the 1-deoxynojirimycin content in mulberry leaves using method of reversed-phase high performance liquid chromatography.The results showed that increment of the concentrations of fructose and inorganic salt,and decrement of phosphate concentration in MS were propitious to the synthesis and accumulation of the 1-DNJ in mulberry leaf.The Fe2+ concentration of medium had a significant influence on the 1-DNJ content.The 1-DNJ couldn't be synthesized if the medium is absent of Fe2+.But the Fe2+ concentration increased from 0.05 to 0.20 mmol/L,the 1-DNJ content was reduced significantly.The existence of Aicotinic acid and VB6in the medium inhibited the synthesis of the 1-DNJ,while the Mg2+,inosital,VB1 and glycin had no significant effect.
