摘    要:目的]探讨百合不同品种和组织中蛋白质的表达特征及其生物学功能。方法]以"西伯利亚"、"索邦"、"黄天霸"3个百合品种为试验材料,采用一维凝胶电泳-液相色谱-质谱技术,对不同品种与组织的差异表达蛋白质组分进行分析与鉴定。结果]SDS-PAGE图谱显示,百合叶片中蛋白质组分最为丰富,其不同组分间的表达量差异也最大,花丝中差异最小,花瓣、花药、柱头、子房等介于两者之间;在3个百合品种之间,检测到6个蛋白质条带的表达量存在差异。质谱鉴定结果表明,这些差异条带共包含了28个蛋白质组分,分别参与叶绿素结合、光系统组成、光合碳固定、糖代谢、蛋白质合成与修饰(磷酸化、糖基化)等生理过程。结论]上述差异蛋白质组分的分离与鉴定,为进一步研究百合遗传特性的分子机理、挖掘其功能基因等提供了一定的试验基础。

Analysis and Identification of Differential Expression Proteins in Different Varieties and Tissues of Lily

Abstract:Objective] The research aimed to study the expression characteristic and biological function of differential proteins in different varieties and tissues of Lily.Method] One dimensional electrophoresis/ liquid chromatography-mass spectrometry were used to analyze and identify on differential expressed proteins from six tissues and three varieties of Lily("Siberia","Sorbonne" and "Manissa").Results] The SDS-PAGE maps displayed that the protein components of Lily leaves were the most abundant and also showed the maximum differences among different components,filaments take second place,and petals,anthers,stigma and ovary were in between.Six bands of differential expressed proteins were detected in the varieties of Lily.The results of mass spectrometric identification showed that these differential expressed protein bands contain 28 protein components which take part in the physiological processes of chlorophyll binding,photosystem,photosynthetic carbon fixation,glucose metabolism,protein synthesis and modification(phosphorylation and glycosylation) etc.Conclusion] The isolation and identification of the differential proteins above provide experimental basis for further study on molecular mechanism of Lily genetic characteristics,searching for new functional gene and so on.
