摘    要:目的]构建毛竹萌发种子的全长cDNA文库。方法]以毛竹萌发种子为材料,利用Oligo-capping法构建其全长cDNA文库。结果]试验得到文库的库容达650万克隆,空载较少;插入片段大小在0.3~5.0 kb之间,片段大小的分级严格且一致性良好,其中显著的是长片段全长cDNA(1.0~5.0 kb)的比例高达30%,达到了高质量全长cDNA文库的标准。结论]毛竹萌发种子全长cDNA文库的成功构建将为竹类植物功能基因组研究奠定重要的前期科研基础。

Construction of Full-length cDNA Library for Germinating Seeds of Phyllostachys heterocycla

Abstract:Objective] This study aimed to construct the full-length cDNA library for germinating seeds of Phyllostachys heterocycla.Method] Germinating seeds of P.heterocycla were used as experimental materials to construct the full-length cDNA library by using Oligo-capping method.Result] The constructed library has a capacity of 6.5×106 recombinant clones in total,and a low proportion of clones without inserted fragmenst;the size of inserted fragments ranges between 0.3-5.0 kb,with strict classification and ideal consistency.Furthermore,the proportion of clones with long inserted fragments(1.0-5.0 kb) was as high as 30%,achieving the standard for high-quality full-length cDNA library.Conclusion] The full-length cDNA library for germinating seeds of P.heterocycla was successfully constructed,which laid important foundation for the functional genomics research of bamboo plants.
