高羊茅种子愈伤组织诱导及植株再生研究

摘    要:该文以MS培养基为基本培养基 ,两个品种的高羊茅种子为外植体 ,接种在附加 2 ,4 D、KT、6 BA、NAA、IAA、水解乳蛋白等不同激素和营养物质的培养基上 ,研究了诱导高羊茅种子愈伤组织形成的主要因素 .结果发现在不同种类和浓度的激素条件下 ,愈伤组织的诱导率有很大的差异 ,2 ,4 D是诱导高羊茅种子形成愈伤组织的关键因素 ,而KT、6 BA、NAA、IAA则没有明显的效果 ,并从多种培养基组合中筛选出了诱导愈伤组织的最佳培养基 ,高羊茅种子的愈伤组织诱导以MS + 6mg L 2 ,4 D + 50 0mg L水解乳蛋白为最佳 .通过愈伤组织的继代和分化培养 ,诱导出丛生芽 ,分化培养基以MS + 4mg L 2 ,4 D + 0 .2mg LKT为最佳 ,初步建立起高羊茅的植株再生体系

Callus induction and plant regeneration from seeds of Festuca arundinacea

Abstract:In this paper, two species of tall fescue seed were cultivated on the MS medium supplemented with different kinds of hormones and nutrients. The effect of 2,4--D,KT, 6--BA, NAA,IAA,lactalbumin hadroly satz and culture c onditions for inducing callus from Festuca arundinacea seeds were described . The frequency of callus induction was different when the same kind of Festuca arundinacea seeds was cultivated in MS medium supplemented w ith different types of and different concentrations of hormones. It was shown that 2,4--D decided the frequency of callus induced from t all fescue seeds, while KT, 6-- BA,NAA,IAA had no significant effect on the callus. The optimum callus in ducing medium of Festuca arundinacea seeds was selected from these kinds of media,and the best one was MS 6 mg/L 2,4--D 600 mg/L l actalbumin hadrolysatz. The authors successfully regenerate tall fescue plant by means of cultivating tall fes cue callus on the MS medium supplemented with 2,4--D and KT,a nd th e best plant regeneration medium of tall fescue was MS 6 mg/L 2,4--D 0.2 mg/L KT. A preliminary plant regeneration system of Festuca ar undinacea was established on the MS medium supplemented with appropriate hormones and nutrients by means of inducing callus and regenerating plants.
