超高效液相色谱法测定鸡蛋中6种色素残留量的方法研究

摘    要:为快速有效地测定鸡蛋中苏丹红Ⅰ、苏丹红Ⅱ、苏丹红Ⅲ、苏丹红Ⅳ、斑蝥黄和β-阿朴-8′-胡萝卜素酸乙酯残留量,将均质混匀后的鸡蛋用乙腈超声提取,氮吹浓缩后乙腈复溶,外标法定量,建立了超高效液相色谱法。结果显示:苏丹红Ⅰ、苏丹红Ⅱ、苏丹红Ⅲ、苏丹红Ⅳ、斑蝥黄和β-阿朴-8′-胡萝卜素酸乙酯在10~10000 ng/mL浓度范围内线性关系良好,相关系数>0.999;检测限为10μg/kg,定量限为20μg/kg;6种色素在4个添加水平的平均回收率为80.3%~97.0%,日内相对标准偏差(RSD)为2.02%~10.91%,日间相对标准偏差(RSD)为1.10%~8.76%。建立的方法简便、快速、高效,适用于鸡蛋中苏丹红Ⅰ、苏丹红Ⅱ、苏丹红Ⅲ、苏丹红Ⅳ、斑蝥黄和β-阿朴-8′-胡萝卜素酸乙酯残留量的测定。

Abstract:In order to determine the residues of Sudan Ⅰ, Sudan Ⅱ, Sudan Ⅲ, Sudan Ⅳ, canthaxanthin and ethyl 8’-apo-β-caroten-8’-oate in eggs, an ultra performance liquid chromatography(UPLC) method was established. The samplewas extracted by acetonitrile, and nitrile was redissolved after nitrogen blowing and concentration, then was quantifiedby the method of external standard. The results showed that the calibration curves of the 6 pigments were good linearrelativities within the concentrations of 10 to 1 000 ng/m L and the correlation coefficient were all more than were allmore than 0.999. The limits of detection was 10 μg/kg, and the limit of qantitation for this method was 20 μg/kg. Theaverage recoveries at spiked concentrations of 20, 40, 200, 2 000 μg/kg ranged from 80.3% to 97.0%, with the relativestandard deviation of inter-day was in the range of 2.02% to 10.91%, and the relative standard deviation of intra-daywas in the range of 1.10% to 8.76%. The established method was simple, quick and high sensitive, and could be appliedto detect Sudan Ⅰ, Sudan Ⅱ, Sudan Ⅲ, Sudan Ⅳ, canthaxanthin and ethyl 8’-apo-β-caroten-8’-oate residues in eggs.
