谷氨酸对热损伤牛小肠上皮细胞抗氧化指标及炎性因子的影响

摘    要:为了研究谷氨酸(Glu)对热损伤奶牛小肠上皮细胞氧化指标及炎性因子的影响,将状态良好的传代至3代的奶牛小肠上皮细胞(Intestine Epithelium Cell,IEC)于42℃培养6 h,造成热损伤模型,在Glu浓度分别为4、8、12、16、20、28、36 mmol/L的细胞培养液中进行培养。12 h后,把所有处理的细胞培养液上清液和细胞全部分别收集保存,检测培养上清液中的抗氧化指标和细胞中炎性因子的基因表达。结果表明:在Glu浓度为8、12、16、20、28、36 mmol/L时,细胞上清液中谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活力显著升高(P<0.05);乳酸脱氢酶(LDH)和丙二醛(MDA)含量与对照组相比呈显著降低的趋势(P<0.05);Glu在低浓度时能够调低促炎因子IL-1β基因mRNA的相对表达量,但在高浓度28、36 mmol/L时,反而调高了IL-1β基因m RNA的相对表达量。在Glu浓度为28 mmol/L时,抗炎性因子IL-10 mRNA的相对表达量高于热损伤组,其他浓度条件下均低于热损伤组。因此,Glu能提高IEC抗氧化能力,降低细胞炎性因子,从而对热损伤IEC具有一定的修复和保护作用。

Effect of Glu on antioxidation and inflammatory factors of bovine intestinal epithelial cells with heat damage
Abstract:To study the effect of Glu on antioxidation and inflammatory factors induced by heat damaged, the intestinal epithelial cells, which in good condition(the third generation), were cultured at 42 ℃ for 6 h, to made a heat damaged modle. The cells were cultured with different concentrations of Glu of 4, 8, 12, 16, 20, 28, 36 mmol/L, respectively. After 12 h, cell supernatants and cells were collected to detect antioxidant index levels and inflammatory factor. The results showed that the activity of GSH-Px and SOD in supernatant was increased significantly at the Glu concentration of 8, 12, 16, 20, 28, 36 mmol/L(P<0.05), the LDH and MDA content decreased significantly compared with the control group(P<0.05), and the relative expression of the pro-inflammatory factor IL-1β gene mRNA was reduced at low concentration, but increased at high concentration of 28, 36 mmol/L. The relative expression of IL-10 gene mRNA, an inflammatory inhibitory factor, was higher than that of heat damaged group only at the Glu concentration of 28 mmol/L, and was lower than that of heat injury group under other concentration conditions. Therefore, Glu can improve the antioxidant capacity of IEC and reduce the cytokines, which can repair and protect the IEC of heat damage.
