摘    要:将小球藻和盐藻在含终浓度为0.5μmol/L的石油培养液中连续培养21 d,利用血球计数法、紫外分光光度法、考马斯亮蓝法和蒽酮比色法,分别测定其生长曲线、叶绿素a含量、胞内外蛋白和胞内外多糖含量。结果显示:2种微藻的细胞密度均在培养第21天达最大值,小球藻生长受到抑制,盐藻与对照组无明显差别;小球藻叶绿素a含量为对照组的48%,而盐藻叶绿素a含量却明显增加,培养第21天是对照组的2.5倍;2种微藻的胞内外蛋白含量均高于对照组,均在培养第10天达最大值,随后逐渐降低,但降低幅度和速度均低于对照组;2种微藻的胞内外多糖含量均低于对照组。研究表明,在含低浓度石油的海水培养液中,小球藻和盐藻都具较好的耐受性,且盐藻比小球藻具有更强的生长能力,更适合在含低浓度石油的海水中大规模培养,具有更高的开发价值。该研究为利用石油污染海水大规模培养微藻的藻种开发奠定了理论基础。

Biochemical Analysis of Chlorella sp.and Dunaliella salina Cultured in Oil Pollution Seawater

Abstract:Chlorella sp. and Dunaliella salina were cultured for 21 days in seawater culture medium with 0.5 μmol/L petroleum. Their growth curve , chlorophyll a content , intracellular and extracellular proteins , intracellular and extracellular polysaccharide content were measured by using blood counting , UV spectrophotometry , coomassie brilliant blue and anthrone colorimetry , development of large-scale cultivation of microalgae. The results showed that the cell density of the two microalgae were up to a maximum on the cultured 21st day , but Chlorella sp. growth was inhibited and Dunaliella salina had no significant difference from the control group. The content of Chlorella sp. chlorophyll a was 48% of the control , however , the content of Dunaliella salina chlorophyll a was significantly increased 2.5 fold compared to the control group. Their content of both intracellular and extracellular protein was higher than control , up to the maximum on the 10th day in culture , and then gradually decreased , but the reducing magnitude and speed were lower than control. Their content of intracellular and extracellular polysaccharide were lower than control group. All these have suggested that both Chlorella sp. and Dunaliella salina were of tolerability when cultured in seawater culture medium containing petroleum of low concentrations. At the same time Dunaliella salina growth was stronger than Chlorella sp. , which was more suitable for large-scale cultivation in seawater containing petroleum of low concentration.
