摘    要:马腺疫是一种高度接触性疫病,困扰着发达国家的养马业。通过筛选马链球菌CF32株3kb~8kb随机基因组文库,获得一株阳性克隆。测序拯救质粒得到序列,确定插入片段编码完整aa的分子量为39.612Ku(Se39.612),根据序列设计特异性引物PCR扩增该基因,分子克隆表达该蛋白。用纯化Se39,612蛋白检测马腺疫康复血清及鼻腔冲洗液,表明Se39,612在体液免疫及粘膜免疫中起作用。

Abstract:Equine strangles is a highly contagious disease which baffled the horse industry in the developed countries.We have identified a novel protein designated Se39.612 by screening a phage library of 3 kb-8 kb random DNA fragments of Streptococcus equi CF32.The inserted segment encoding the entire amino acid sequence of Se39.612 was determined by sequencing the rescued plasmids.Se39.612 gene was amplified using specific primers and cloned,and protein was expressed and purified.Strong reactivity of the protein with convalescent sera(IgG) and mucosal wash(IgA) was detected during infection,suggesting its role of humoral and mucosal responses.
