| 摘 要： | 采用对虾白斑综合症病毒和传染性皮下组织坏死病毒的特异引物,在一个PCR反应中同时扩增到大小分别为271 bp和356 bp的病毒特异片段,多重PCR条件优化后可从最少100 fg级病毒DNA模板中定性检测出此两种病毒。

| Abstract: | A method of detecting white spot syndrome virus(WSSV) and infectious hypodermal and haematopoietic necrosis virus(IHHNV) in shrimps was developed by using a single-tube,one-step mutiplex polymerase chain reaction(PCR).Four special primers were prepared,based on the analysis of WSSV and IHHNV DNA sequence.It was shown that all samples containing WSSV and IHHNV were amplified by the mutiplex PCR,yielding two specific bands of WSSV 271 bp and IHHNV 356 bp,but no specific band was amplified from other shrimp virus.As small as 100 fg WSSV and IHHNV DNA were detected using gel electrophoresis in this mutiplex PCR. |
