猪瘟病毒与猪流行性乙型脑炎病毒复合RT-PCR检测方法的建立

摘    要:猪瘟病毒(CSFV)和流行性乙型脑炎病毒(JEV)是引起种猪繁殖障碍的病原,而且经常混合感染,及时准确诊断是防治的前提.根据CSFV的E2基因保守序列和JEV的E基因保守序列,选取 2 对引物建立检测CSFV、JEV的复合RT-PCR方法,分别扩增出了大小为 508 和1 015 bp 特异性基因片段.应用此方法检测了10份临床样品(淋巴结、脾、肝、肺等组织),结果4份CSFV阳性,3份JEV阳性,其余为阴性.结果表明此CSFV 和 JEV 复合RT-PCR 诊断方法,具有很好的特异性和敏感性,可用于临床检测.

Abstract:Classical swine fever virus(CSFV) and Japanese encephalitis virus(JEV) are the major causes of some serious reproductive diseases and confection is common.Therefore rapid and reliable diagnosis is crucial for effective prevention and treatment.In this study,a multiplex RTCR was developed using two-pairs primers designed according to conserved sequences in E2 fragments of CSFV and that in E fragments of J EV.PCR products of 508bp and 1O15bp were obtainedfor respective viruses.Ten organ samples(lung,lumph,liver,spleen,etal) were respectively detected by the multiplex RT-PCR,out of which four were CSFV positive,three were JEV positive,the control samples were negative.The results showed that the multiplex RT-PCR of CSFV andJEV was a rapidly specific and sensitive diagnostic method.
