摘    要: 利用基因枪法, 以豇豆(V igna aconitifolia) 的Δ1 - 吡咯啉- 5 - 羧酸合成酶基因( P5CS ) 的突变型P5CS-F129A基因转化高羊茅的下胚轴愈伤组织, 获得25株经PCR检测和Southern blot分析为阳性的转基因植株, 转化频率约为1.4%。脯氨酸含量测定表明, 转基因植株的脯氨酸含量比对照高31% ~83%。对植株的抗旱性初步检测表明, 转基因植株比对照耐旱。

Abstract:In this study, a mutated form of V igna aconitifolia Δ1-pyrroline-5-carboxylate synthetase( P5CS -F129A ) gene was transferred into calli of tall fescue by micro p rojectile bombardment. Twenty2five transgenic p lants were obtained through phosphinothricin screening, PCR and Southern blot analysis. Thetransformation frequency in calli of tall fescue was about 1.4%. The content of proline in transgenic plants was 31% - 83% higher than that in untransformed plants. Furthermore, the transgenic p lants were more tolerant to drought stress.
