大叶樟无菌芽诱导因素研究

摘    要:为获得大叶樟种苗无菌芽,构建大叶樟遗传转化体系。以大叶樟茎段为材料,研究不同季节、不同树龄、不同芽类型和不同激素配方对外植体萌芽的影响。结果表明,大叶樟外植体在6-BA含量和6-BA/NAA比值越高的培养基上萌芽率越高;幼年树外植体的萌芽率高于成年树,染菌率低于成年树;外植体中顶芽的萌芽率低于腋芽,污染率高于腋芽;外植体在春季的萌芽率高于秋季,污染率低于秋季。结论,大叶樟无菌芽诱导最好在春季采取幼年树上带有腋芽的茎段为外植体,培养基中激素配方为2.0 mg/L 6-BA和0.02 mg/L NAA。

Study on inducing factors of aseptic bud of Cinnamomum septentrionale

Abstract:The genetic transformation system was established to obtain sterile buds of Cinnamomum septentrionale seedlings. The effects of different seasons, different tree ages, different bud types and different hormone formulations on the germination of explants were studied. The results showed that the germination rate of Cinnamomum septentrionale explants was higher when the content of 6-BA and the ratio of 6-BA/NAA were higher. The germination rate of young explants was higher than that of adult trees, and the bacterial contamination rate was lower than that of adult trees. The germination rate of terminal bud in explants was lower than axillary bud, but the contamination rate was higher than axillary bud. The germination rate of explants in spring was higher than that in autumn, and the contamination rate was lower than that in autumn. Conclusion: In spring, it is best to use stem segments with axillary buds in young trees as explants for germ-free bud induction. The hormone formula in culture medium is 2.0 mg/L 6-BA and 0.02 mg/L NAA.
