葛根素对镉致大鼠血脑屏障损伤的保护效应

摘    要:为探究葛根素在镉暴露引起大鼠血脑屏障损伤中的保护效应,24只雄性SD大鼠随机分为4组,分别为对照组、葛根素组、镉组、镉与葛根素共处理组。试验期间,每天按200 mg/kg剂量的葛根素(溶解于5 g/L羧甲基纤维素钠)对葛根素组、镉与葛根素共处理组大鼠进行灌胃,同时以相同剂量的5 g/L羧甲基纤维素钠将对照组、镉组大鼠进行灌胃;14 d后,镉组、镉与葛根素共处理组大鼠在灌胃的同时自由饮用镉水(50 mg/L),持续90 d。试验结束后,分离大鼠大脑皮质,采用荧光定量PCR(qRT-PCR)检测神经型一氧化氮合酶(nNOS)、诱导型一氧化氮合酶(iNOS)、基质金属蛋白酶9(MMP-9)基因表达量,微板法测定一氧化氮(NO)含量,Western blot检测MMP-9、白蛋白(ALB)表达水平,免疫组化染色检测ALB分布情况。结果显示:和对照组相比,镉组nNOS、iNOS mRNA水平,NO含量,MMP-9 mRNA和蛋白水平,ALB蛋白水平极显著升高(P<0.01);与镉组比较,镉与葛根素共处理组nNOS、MMP-9 mRAN水平,NO含量极显著下降(P<0.01),iNOS...

Abstract:In order to explore the blood-brain barrier disruption induced by cadmium(Cd) exposure in rats and the protective effects of puerarin(Pur) in the case, twenty-four male SD rats were randomly divided into four groups: the control group, the Pur group, the Cd group, and the Cd and Pur co-treated group. During the experiment, when Pur(200 mg/kg, dissolved in 5 g/L Carboxymethylcellulose sodium) was given to the rats by gavage in the Pur group and the co-treated group, the same volume of 5 g/L Carboxymethylcellulose sodium was given to the rats by gavage concurrently in the control group and the Cd group. Fourteen days later, the rats in the Cd group and the co-treated group received drinking water containing 50 mg/L Cd plus the gavage for 90 days. After the experiment, the cerebral cortex was isolated from the rodents. The mRNA levels of their neuronal nitric oxide synthase(nNOS), inducible nitric oxide synthase(iNOS) and matrix metalloproteinase 9(MMP-9) were analyzed using qRT-PCR. The contents of nitric oxide(NO) in them were determined using the microwell plate method. The expressions of MMP-9 and Albumin(ALB) in them were detected by Western blot. The distribution of ALB was detected by immunohistochemical staining. The results showed that Cd-induced upregulation of iNOS mRNA, MMP-9, and ALB in the rats were significantly inhibited by Pur(P<0.05). Cd-induced increase of nNOS, MMP-9 mRNA, and NO contents in them were significantly abated by Pur(P<0.01). The above results indicated that Pur did possess a protective effect on the blood-brain barrier disruption caused by Cd in rats.
