| 摘 要： | 从石斛兰病株中获得一个病毒分离物，利用曼陀罗进行分离纯化和繁殖，并通过梯度离心获得提纯病毒，经生物学、血清学鉴定及电镜观察，鉴定该分离物属于建兰叶病毒。分别用纯化病毒及ＳＤＳ－ＰＡＧＥ回收病毒外壳蛋白免疫家兔。获得２种特异性抗血清，并用ＳＰＡ－ＥＬＩＳＡ方法对兰花样品进行了检测研究。

| Abstract: | A virus isolate was obtained from a Dendrobium plant in Hainan through sap inocula-tion on Datura stramonium, an indicator plant. Single lesion inoculati0n was made on Datura stramonium more than three times. The isolate was purified with the procedure lncluding differ-ential centrifugation and Cs2SO4 density gradient centrifugation. Biological, serological and IEM tests showed that this isolate was Cymbidium mosaic virus. The purified virus and the coat pro- tein of the virus collected from SDS-PAGE were used separately to immunize rabbits for prepara-tion of antiserums. Two specific antiserums were obtained and then employed for detection of the orchids and vanillas through SPAEI,ISA. |
