摘    要:以膜荚黄芪的嫩叶片为材料,对CTAB法和高盐低pH法进行了改良,并用此改良法提取总DNA,用紫外分光光度法和琼脂糖凝胶电泳检测DNA质量。结果表明,无论从提取率还是纯度上,优化的高盐低pH法都优于CTAB法,但高盐低pH法提取的DNA不适合长期保存。

Abstract:In this paper, the CTAB method and high-salt low-pH method was modified and improved to extract total DNA using the young leaves of Astragalus membranaceus. Then the DNA quality was detected by UV spectrophotometry and agarose gel electrophoresis. The results showed that the optimization of high-salt low-pH method was superior to CTAB method in terms of extraction rate or purity, but the extracted DNA was not suitable for long-term preservation.
