摘    要:目的]优化红景天中红景天苷的提取工艺。方法]用密闭升温产生的高压提取红景天根中的红景天苷,高压液相色谱法测定红景天苷的含量,单因素试验选择提取溶剂,正交试验确定最佳提取工艺,并与超声波法进行比较。结果]提取溶剂为水时,红景天苷的得率最高。提取时间对红景天苷得率的影响最大,其次是提取温度,两者为决定性因素,然后是料液比、浸泡时间。高压法最佳提取工艺条件下红景天苷的得率高于超声波法,避免了超声波及微波对人体的危害,成本低,更适用于工业化生产。结论]水作提取溶剂时,高压法提取红景天苷的最佳工艺为:料液比1∶16(g/ml),浸泡时间1.0 h,提取温度140℃,提取时间30 min。

Abstract:Objective ] The aim of the research was to optimize the extraction technology of salidroside from Rhodiola rosea L.Method] Salidroside was extracted from the root of R.rosea by high pressure produced by the closed heating. Salidroside content was determined by high pressure liquid chromatography.Solvent was selected through single factor test.The optimum extraction technology was confirmed through the orthogonal test and it was compared with that by ultrasonic method.Result] When water was taken as extraction solvent,the yield of salidroside was highest.The effect of extraction time on the yield of salidroside was greatest,followed by extraction temperature,and the next were material-liquid ratio and soaking time.Extraction time and extraction temperature were the decisive factors.The yield of salidroside under the optimum extraction technology by high pressure method was higher than that by microwave method.It avoided the harm of ultrasonic and microwave to human body.Its cost was low and it was more suitable for industrialized production.Conclusion] When the water was taken as extraction solvent,the optimum extraction technology of salidroside by high pressure was as follows: the material-liquid ratio of 1∶16,soaking time of 1.0 h,extraction temperature of 140 ℃,the extraction time of 30 min.
