摘    要:通过对台湾山苏蕨不同外植体的离体培养,探讨适宜台湾山苏蕨离体培养的最佳外植体材料,建立无菌培养体系,筛选适宜的诱导培养基、增殖培养基、生根培养基及移栽基质配方。结果表明:孢子囊是最佳外植体,在MS及1/2 MS诱导培养基中诱导率高达464% 最佳增殖培养基为MS+6-BA 0.3 mg/L+NAA 0.1 mg/L或MS+KT 0.9 mg/L+NAA 0.1 mg/L,增殖率可达4.3以上 最佳生根培养基为1/2 MS+NAA 0.5 mg/L(或ABT 0.5 mg/L),生根率可达95%以上。生根试管苗在纯黄心土与细沙(8∶2)和黄心土与泥炭土(8∶2)2种基质中,移栽成活率均可达98%以上。

Abstract:A tissue culture experiment was conducted with varied explants of Asplenium nidus in order to explore the optimal explant materials, screen proper induction medium, proliferation medium, rooting medium and transplant medium formulation, and to-set up sterile culture system. The results showed that sporangium was the optimal explant and its induction rate on MS medium and 1/2MS medium was up to 464%. The optimal formulation of proliferation medium was MS +6- BA 0. 3 mg/L + NAA 0. 1 mg/L or MS + KT 0. 9 mg/L + NAA 0. 1 mg/L, with proliferation rate over 4.3. The optimal formulation of rooting medium was 1/2 MS + NAA 0.5 mg/L ( or ABT O. 5 rag/L), on which the rooting rate was up to 95%. The survival rate after transplanting was over 98% when the rooted tube-seedlings were transplanted into the pure yellow subsoil with fine sand (in proportion of 8:2) and the yellow subsoil with peat soil (in proportion of 8:2).
