银杏叶总黄酮提取工艺的正交设计研究

摘    要:为了筛选银杏叶总黄酮提取最佳工艺,试验运用正交设计法,考察了溶剂浓度、料液比、提取次数和提取时间4个因素对银杏叶总黄酮提取工艺的影响,并用紫外吸收法测定银杏叶总黄酮的含量。研究结果表明:芦丁标准品在0.05~0.30 mg.mL-1范围内,线性良好(r=0.9920),该提取方法操作简便,结果准确可靠。以总黄酮含量为指标,确定了60%乙醇,料液比1∶8,提取2次,每次1h为最佳提取条件。

Abstract:To optimize the extraction of total flavanoid from leaves of ginkgo bilobal,we used the four factors,i.e.the concentration of ethanol,volume of solvent,number of extraction and extraction time.The effect of the four factors on the concentration of total flavanoid in leaves of ginkgo bilobal is examined by orthogonal design and ultraviolet spectrophotometric technique.The result indicated that the linear relationship of rutin sample was good(r=0.9920) in the range of the 0.05 ~ 0.3 mg·mL-1.60% ethanol,1∶8,2 times,1h is the best extraction method.The method is simple,accurate and reliable.
