利用分子生物学技术鉴别真、假杂交稻的研究

摘    要:对由湖北省某公证处封存的两份杂交稻材料进行DNA、蛋白质指纹鉴定,首先采用业已建立的可特异鉴定以明恢63为父本的杂交稻真、假的P18引物进行RAPD分析,发现两份材料的叶片及种子DNA在0.8kb扩增带上存在着有和无的区别;而所用的42个水稻微卫星标记,发现5个水稻微卫星标记具有多态性;在此基础上,进一步对两样品的种子水溶性蛋白质毛细管电泳图谱进行比较,二者在出峰时间及峰值上存在差异。上述结果表明,两份材料并非同一杂交组合,但在遗传背景上存在较大的相似性。

Abstract:In this study, two hybrid rice samples from Hubei province were analyzed to identify true or false hybrids with DNA markers and Protein Capillary Electrophoresis. A primer P18 was used to amplify their DNAs. Results showed that one sample of seeds and their seedlings had a 0.8 kb special band, and the other without the band. Then, 42 rice microsatellite markers were used to amplify the two samples, as a result, 5 polymorphic microsatellite markers were found among those markers. Furthermore, the capillary electrophoresis was applied for assaying the two samples. Two different chromatography was obtained. The results of DNA and protein fingerprinting indicated that the two samples were not from the same hybrid combination, but with genetic background similarity.
