| 摘 要： | 本试验用DNA池法从40条随机引物中筛选出29条多态性、重复性好的引物,对贵州黄鸡、兴黔黄鸡、乌骨鸡三个培育品种(系)和地方品种兴义矮脚鸡及引进选育品种矮脚黄鸡共五个品种(系)进行随机扩增多态亲缘关系分析。结果表明,29条引物共扩增出稳定、清晰、重复性好的条带190条,其中多态性片段127条,在300bp￣3000bp之间。单个引物扩增出的RAPD条带在4￣13条之间,平均为6.55条。五个品种(系)间的亲缘关系符合它们的育种背景1,2,3]。

| Abstract: | Randomly amplified polymorphic DNA (RAPD) was used to investigate the blood relationship of five varieties (lines) of chicken including Guizhou brown chicken, Xingqian brown chicken, Xingyi bantam chicken, yellow bantam chicken and black bone chicken. Twenty nine primers with better polymorphism and repeatability were selected out from the total 40 ones using technique of DNA pool. Fine polymorphism was detected within the five varieties (lines) of chicken using these selected 29 primers. One hundred and twenty seven polymorphic bands from total 190 ones were detected with molecular weight between 300 bp and 3000 bp according to standard DNA marker. The bands amplified by single primer are between 4 and 13, averaged 6.55. The blood relationship investigated by randomly amplified polymorphic DNA is in consistence to that elucidated by traditional breeding methods. |
