摘    要:利用SSR分子标记技术对12份蕨类材料进行亲缘关系分析。结果表明,从180对引物中筛选出15对多态性高、重复性好的引物,对12份蕨类材料基因组DNA进行扩增,共扩增出117条带,其中多态性带98条,平均每对引物产生6.53条多态性带,多态性比例为83.76%。通过NTSYS—pc2.10e软件计算不同蕨类材料之间的遗传相似系数,并进行聚类分析。遗传分析表明,12份材料间的遗传相似系数为0.351~0.857,且聚类分析表明,遗传相似系数在0.61—0.63可将供试材料分为3个类群。应用SSR分子标记技术能较准确地分析蕨类不同材料之间的亲缘关系及遗传多样性。

Analysis of Genetic Relationship of 12 Pteridophyte Cultivars by Using SSR Markers

Abstract:In this study, the molecular marker system SSR (simple sequence repeat) was used to analyze the genetic relation ship of 12 pteridophyte cultivars. The results showed that 15 pairs of primers with high polymorphism and good repeatability were screened out from 180 pairs of primers, and they were used to amplify the genomic DNA of these pteridophyte cuhivars. A total of 117 bands were amplified, and 98 of them were polymorphic, with an average of 6.53 polymorphic bands per primer pair, with the polymorphism rate of 83.76%. The genetic similarity coefficients were calculated and the cluster analysis was conducted by using NTSYS pc 2.10e software. The genetic analysis showed that the genetic similarity coefficients between the 12 cuhivars ranged from 0. 351 to 0. 857. The cluster analysis indicated that all the cuhivars could be categorized into three major clusters. The genetic rela tionship and genetic diversity of pteridophyte materials could be assessed efficiently by SSR markers.
