光和糖对非洲菊花色素苷积累及CHS、DFR 基因表达的影响

摘    要: 探讨了光和糖对非洲菊(Gerbera hybrida) 花序最外轮舌状花( ray floret) 花瓣着色的影响。P2期花序和舌状花先在含有3%蔗糖和无蔗糖的固体培养基(8 g·L-1琼脂) 上进行3 d暗培养, 光照处理不同时间后, 继续暗培养, 第9 天取样, 采用分光光度计法测定舌状花花瓣花色素苷积累; 照光后立即取样, 以Northern-blot检测CHS、DFR基因表达的变化。研究结果表明, 短暂光照提高花色素苷积累; 光照明显诱导CHS、DFR基因表达, 光照时间越长, 基因表达水平也越高。暗培养7 d和9 d, 光诱导基因表达效应明显减弱。培养基中含有蔗糖时, 光强烈诱导花色素苷积累和CHS、DFR基因表达。

Abstract:Effects of light and sugar on Gerbera hybrida ray floret ( rf) p igmentation were investigated.Inflorescences or rfs at stage P2 were detached and incubated on the medium (8 g·L-1 agarwith 3% sucroseor free of sucrose) under 14 h light /10 h dark cycles at 23 - 25℃. Cultured inflorescences and rfswere firstincubated in darkness for 3 d, then exposed to transient light for a period of time followed by an incubation inthe dark until harvesting for anthocyanin determination by spectrophotometer at 9 th d or harvested immediatelyfor gene exp ression detected byNorthern-blot. The results showed that transient light exposure induced anthocyanin accumulation and the gene exp ression of CHS and DFR in rf petal after 3 d p reculture in darkness. Thelonger time of light irradiation strongly enhanced the gene exp ression of CHS and DFR . Petals precultured indarkness for 7 d and 9 d had weak response to light in gene expression. When sucrose was present in the medium, both anthocyanin accumulation and gene exp ression were strongly increased under light condition.
