| 摘 要： | 家蚕后丝腺和脂肪体总RNA含量随着五龄期的增长而增加,在五龄中期达到最高水平.用Sepharose2B柱层析分离RNA各组分,观察到家蚕后丝腺rRNA,tRNA和mRNA的含量在五龄期的变化均与总RNA相对应.JHA ZR515处理家蚕后,后丝腺的RNA及其组分rRNA.tRNA和mRNA均明显增加.脂肪体RNA也有增加.这些结果显示出JHA增丝的原因可能与JHA在转录和翻译水平上促进了丝素蛋白基因的表达有关.

| Abstract: | The RNA synthesis in the posterior silkgland and fat body of Bombyx mori increases with the duration of the 5th instar go on, and reaches a maximum level in the mid-5th instar, and then decreases slightly. By using Sepharose 2B column chromatography RNA components were separated and assayed, it was observed that fibroin mRNA and rRNA in posterior silkgland cells of the silkworm exhibit almost the same changes, and corresponding to the total RNA. During the last half of the 5th instar the mRNA and rRNA comprise from 4.5% to 6.5% and from 78% to 85% respectively, of the total RNA by weight.
The application of JHA ZR515 in the early 5th instar larvae of the silkworm leads to the longer duration of the instar and to a 20% increase of cocoon shell weight. In JHA-treated larvae the contents of RNA and its components, rRNA and mRNA, from the posterior silkgland cells were markely increased respectively than that of the controls; The RNA contents in the fat body cells were also higher than the controls.
These results indicated that the mechanism of silk increase by using JHA is not only due to the prolongation of the 5th instar duration of the silkworm but also due to the increase of the contents of RNA and its components,rRNA and fibroin mRNA in the posterior silkgland of the silkworm. |
