| 摘 要： | 为了建立兽用白头翁颗粒中白头翁皂苷B4的含量测定方法.利用HPLC法,采用HypersilC18柱（250 mm×4.6 mm,5μL）,以乙腈—0.1％磷酸（27∶73,V/V）为流动相,检测波长为205 nm,流速1.0 mL/min,柱温30℃.以白头翁皂苷B4对照为参比,在白头翁颗粒色谱图中,对白头翁皂苷B4的保留时间及峰面积的RSD进行分析.采用外标法测定3批白头翁颗粒中白头翁皂苷B4的含量.试验结果显示,白头翁皂苷B4在0.1～2.0 mg/mL范围内呈良好的线性关系,（r=0.9995,n=6）,平均含量为11.29mg/g,平均回收率为99.28％ （RDS=2.09％,n=9）.该方法操作简便、结果准确,重复性好,可用于兽用白头翁颗粒的质量控制.

| Abstract: | To establish a method for determining the content of Pulchinenoside B4 in veterinary Baitouweng granules,the HPLC method was adopted on the Hypersil C18 （250 mm×4.6 mm,5 μm） column.The mobile phase was consisted of acetonitrile-0.1 % phosphoric acid （27：73,v/v） at the flow rate of 1.0mL · min-1,column temperature of 30 ℃ and detection wave-length of 205 μm.The injection volume was 20 μL.In the recorded chromatogram of the test preparation,Pulchinenoside B4 was used as a reference substance,and RSD （%） of the retention time and the peak areas were measured.Using the external standard method,the content of Pulchinenoside B4 was determined in the 3 batches of preparation.The experimental results showed that the linear range of Pulchinenoside B4 was 0.1 ～2.0 mg/mL with average content of 11.29 mg/g,average recovery of 99.28% （RDS=2.09%,n=9）.This method is simple and accurate with a good reproducibility and can be used for the quality control of veterinary Baitouweng granules. |
