摘    要:利用改良的SDS法提取棉花干种子的基因组DNA,所得基因组DNA经分光光度计、琼脂糖凝胶电泳和随机SSR引物扩增效果检测,结果表明利用改良的SDS法直接所提取棉花干种子的基因组DNA,符合棉花SSR-PCR反应对DNA质量的要求。本研究为棉花SSR分析建立了一种快速、简便的干种子DNA提取方法,为SSR标记在棉花遗传育种研究中的应用奠定了基础。

Abstract:In this study,an improved SDS method for DNA extraction from cotton dry seeds was developed.The quality of DNA obtained from 32 cotton cultivars was evaluated in terms of the Eppendorf BioPhotometer analysis,agarose gelelectrophoresis,and SSR-PCR analysis.The results indicated that the extracted genomic DNA was of good quality,and can satisfy the need of SSR-PCR.This rapid and effective DNA extraction method provides a foundation forthe application of SSR in cotton heredity and breeding research.
