黑苦荞黄酮提取工艺及其胶囊制备研究

摘    要:以总黄酮提取率为评价指标,通过苦荞黄酮提取单因素试验和正交试验确定乙醇回流提取苦荞黄酮的最佳工艺条件为苦荞粉粒径100目,乙醇体积分数60%,料液比1∶35(g∶mL),提取温度80℃,提取时间150 min,在此条件下总黄酮提取率达44.68±2.29 mg/g。胶囊成型工艺中,以粉体的吸湿率、颗粒的成型率、休止角和堆密度等为评价指标,确定苦荞黄酮粉与辅料微晶纤维素质量比为1.5∶1,以85%乙醇为润湿剂进行制粒,在此条件下制备的胶囊产品抗潮湿性能强,流动性好,符合生产要求。考查产品临界相对湿度,确定生产环境相对湿度应控制在67.85%以下。根据中国药典方法,对研制产品进行水分含量、装量差异及崩解时限的分析检测,结果显示均符合药典规定。

Abstract:The optimum extraction conditions of flavonoid from black tartary buckwheat were confirmed by single factor experiments and orthogonal experiment as follows:grain size of black tartary buckwheat powder 100 mu,ethanol concentration 60%,ratio of black tartary buckwheat powder to extraction solution 1∶35(g∶mL),extraction temperature 80℃,extraction time 150 min.The extract rate of flavonoid under the optimum extraction conditions was 44.68±2.29 mg/g.The optimum formulation process was confirmed by measuring hygroscopicity,formability,angle of repose and pile density.The extract powder and microcrystalline cellulose were mixed evenly at 1.5∶1.And 85%alcohol was used as the wetting agent.The granules of the end product had a good fluidity and anti-wet performance.The critical relative humidity was about 67.85%.The moisture content,weight difference and disintegration time limited of the end product were analyzed.The results showed that the quality of the black tartary buckwheat hypoglycemic capsule was in line with the Chinese Pharmacopoeia requirements.
