大晶帽石斛再生体系建立研究

摘    要:本文以成熟度70%原生大晶帽石斛荚果内种子为外植体,探究采用不同消毒时间、培养基成分构成及浓度配比对大晶帽石斛无菌萌发的影响。结果表明,大晶帽石斛荚果用5%次氯酸钠消毒15 min,可有效降低污染率;将消毒后荚果种子接入MS+蛋白胨2 g/L+NAA 0.5 mg/L+花宝1号2 g/L+0.1%活性炭+0.58%琼脂+2%蔗糖培养基中,种子萌发率达88%,是适宜大晶帽石斛无菌萌发的培养基配方。

Study on the regeneration system of Dendrobium macrocephalum

Abstract:The pods of Dendrobium macrocap with maturity of 70% were used as explants, the effects of different composition and concentration of medium and disinfection time on the aseptic germination of Dendrobium Dendrobium were investigated in order to explore the most suitable treatment method for the aseptic germination of Dendrobium. The results showed that Disinfect pods with 5% sodium hypochlorite for 15 minutes, It can effectively reduce the pollution rate after inoculation, adding MS+seed peptone 2g.L-1 + NAA 0.5 mg.L-1 + 2 g/L + active charcoal 0.1% + agar 0.58% + sucrose 2% was the best treatment, the seed germination rate can reach 88%.
