摘    要:目的]为研究传染性支气管炎(IBV)的流行变异及其防治奠定基础。方法]从辉县某大型养鸡场疑似IBV的发病鸡群中采集病料,进行病毒分离和鉴定,并对分离病毒的M基因进行RT!PCR扩增。结果]从鸡病料中分离到1株IBV,命名为IBV!HN05。经浓度1%胰酶、卵磷脂酶C和10%乙醚处理后,该病毒尿囊液可以凝集鸡、绵羊、人、小鼠和兔的红细胞,而未处理的病毒尿囊液则无此凝集活性。该分离株IBV-HN05和标准毒株IBVM41对NDVLaSota都有抑制作用。该病毒对9日龄鸡胚的致病变率达100%。通过鸡胚矮小试验和动物回归试验,发现该分离株能引起鸡胚和病鸡典型的临床症状和病理变化。PCR扩增结果进一步证实了该分离株为IBV病毒。结论]该研究为SARS病毒的研究提供了动物模型。

Abstract:Objective] The research aimed to lay the foundation for studying the prevalence and variation of infectious bronchitis virus(IBV)and its control. Method] The diseased materials were collected from the infected chicken flock suspected of IBV in a large-scale chicken farm of Hui county to isolate and identify the virus and RT-PCR amplification was made on M gene in the isolated virus. Result] One strain of IBV was isolated from the diseased chicken materials and named as IBV-HN05. After treating with 1% pancreatin, lecithinase C and 10% aether, the allantoic fluid of this virus could agglutinate the erythrocytes of chicken, sheep, human being, mouse and rabbit, while the allantoic fluid of this virus without this treatment had no this agglutinative activity. Both the isolated strain IBV-HN05 and standard strain IBV M41 had inhibitory effects on strain NDV LaSota. The pathogenic rate of this virus to 9-day-old chicken embryo reached 100%. In the chicken embryo dwarf test and animal regression experiment, it was found that the isolate typical clinical symptoms and pathological changes in chicken embryo and diseased chicken. The results of PCR amplification further proved that the isolated strain was IBV virus. Conclusion] The research provided animal model for studying SARS virus.
