转基因苹果研究进展

摘    要:十多年来,转基因技术以其独特的魅力在苹果上得到了迅速发展。目前,农杆菌介导法是苹果上应用的主要转化方法,叶片再生不定芽途径是被广泛应用的受体系统。影响这一受体系统的主要因素有基因型、外植体的来源、叶片发育阶段及生理年龄、培养条件以及抗生素等。而菌株的类型、侵染时间、共培养的时间和条件、酚类物质及转化植株的选择方式等则影响基因的转化。已有多种标记基因和目的基因转入苹果中,且遗传分析表明外源基因能够稳定遗传。但依然存在转化效率不高、外源基因表达强度不够、可转化的目的基因有限及对转化植株的选择不够重视等诸多问题。综合有关文献对上述问题进行了讨论并对转基因苹果的前景进行了展望。

Abstract:Significant progress of genetic transformation in apples has been made during the past decade.At present,Agrobacterium mediated transformation of leaves are widely adopted in apples,and transgenic shoots were mainly obtained from adventitious bud regeneration.The factors affecting adventitious bud regeneration from leaves include genotype,the source of explants,the development stage and physiological age of leaves,culture conditions and antibiotics;while Agrobacterium strains,infection time,the time and conditions of co-cultivation,hydroxybenzenes and the selection method of transgenic shoots affect the gene transformation efficiency.Hitherto many marker genes and aim genes had been transferred to apples,and expressed stably in progeny.The current problems such as low transformation efficiency and expression,the limited aim genes and less emphasis on the selection of transgenic shoots are discussed and the prospect is put forward in this paper.
