沿海滩涂镉吸附细菌的分离、鉴定及生长特性的研究

摘    要:采用常规细菌分离技术从沿海滩涂的泥样中分离出两株对重金属Cd2+具有吸附作用的细菌(编号为J2和J6),并采用原子吸收法测定了菌株及培养液中Cd2+的含量。结果表明:两株菌在培养液中培养7d后,上清液中Cd2+的含量与对照组相比显著降低(P〈0.05),菌体细胞中Cd2+含量与对照组相比显著增加(P〈0.05);电镜下观测,两株菌均为短杆状,革兰氏染色为阴性,有鞭毛。生理生化特征测定结果显示,两株菌均符合弧菌属的特征,用O/129纸片测试为敏感,TCBS中生长变为黄色,与过氧化氢酶、氧化酶、硝酸盐还原反应为阳性,生长均需要Na+。结合16S rDNA序列分析,J2菌株被鉴定为Vibrio paci-nii,J6菌株被鉴定为溶藻弧菌Vibrio alginnolyticus。J2菌株生长的最适pH为7,温度为20℃,NaCl为30g/L;J6菌株生长的最适pH为7.5,温度为30℃,NaCl为40 g/L。

Abstract:Two Cd^2+ adsorption strains of bacteria J2 and J6 were isolated from coastal beach by general technique, and the adsorption of Cd^2+ was detected by Atomic Absorption Spectroscopy(AAS). The growth and biological characteristics of J2 and J6 were tested to identify the strains. The results showed that they can reduce Cd^2+ consistency in culture medium, while the amount of Cd^2+ in bacterial cells was found to be increased compared with the control (P〈 0.05) 7 days after culture. The J2 and J6 showed gram negative ,rod shape, 0129 paper sensitive, yellow colony in TCBS, catalase positive, oxidase positive, nitrate reduction positive and requirement of Na^+ for growth. The J2 and J6 were identified as genus Vibrio. The sequence of 16S rRNA gene of J2 was found to be accorded with Vibrio pacinii with identity of 99%, and J6 was accorded with Vibrio alginnolyticus with identity of 99%. The optimum growth of J2 was observed under the conditions of pH 7, 20℃, NaCl of 3% and J6 pH 7.5,30℃, and NaCl of 40%.
