枯草芽孢杆菌DPG-01发酵条件的优化

摘    要:通过单因子试验及正交试验对枯草芽孢杆菌DPG!01的培养基组成和发酵工艺条件进行了研究。结果表明,最优化培养基组成为:高温豆饼粉2.0%,尿素0.1%,玉米浆0.15%,玉米淀粉1.00%,磷酸氢二钾0.3%,磷酸二氢钾0.15%,硫酸镁0.1%,硫酸锰0.02%;培养条件为:pH值7.5(消前),装量100ml/500ml三角瓶,摇床频率200r/min,温度(37±1)℃。优化条件下枯草芽孢杆菌DPG!01发酵水平达到9.1×109cfu/ml,芽孢形成率达到90%。

Abstract:Fermentation culture medium and technological conditions of Bacillus subtilis DPG-01 were studied by single factor experiments and orthogonal experiments. The results showed that the optimum medium formula was 2.0 % bean cake, 0.1 % carbamide, 0.15 % corn steep liquor, 1.00 % cornstarch, 0.3 % K2HPO4, 0.15 % KH2PO4, 0.1 % MgSO4·7H2O and 0.02 % MnSO4·H2O. The optimum fermentation conditions were pH 7.5(initial value), amount of load 100 mL/500 mL flask, vibration frequency 200r.p.m and temperature 37±1℃. Under the optimum conditions the fermentation cell density was 9.1×109 CFU/mL with spore ratio of 90 %.
