摘    要:选择拟南芥AtOvKLP蛋白作为研究对象,对其氨基酸序列和结构域进行了分析.结果表明,AtOvKLP蛋白ATP结合位点序列(FAYGQTGSGKT)与大多数动蛋白中的ATP结合位点序列不完全一致(YGQTGS-GKT);通过与动蛋白其它亚家族的系统分析比较,发现AtOvKLP与KHC亚家族类的各动蛋白距离较近;同时构建了pET-khc原核表达载体,并得到了体外表达蛋白.

Abstract:AtOvKLP is one of kinesin-like proteins in Arabidopsis thaliana.The sequencing and motore region analysis displayed ATP-binding sequence of AtOvKLP (FAYGQTGSGKT)was different from other kinesins'(YGQTGSGKT). A close relationship between AtOvKLP and kinesins of KHC subfamily was supported by the phylogenetic analysis among AtOvKLP and other subfamilies. And expressed the AtOvKLP cDNA sequence in E.coil and obtained desired products in vitro for further study in validating the biochemistry activity of AtOvKLP.
