胡杨组培及愈伤组织增殖研究

摘    要:胡杨是我国西北荒漠珍贵先锋树种,但扦插繁殖难以生根。本文以胡杨的枝段为外植体,通过筛选最佳消毒方式和优化培养基,建立胡杨愈伤组织的诱导和增值的培养体系。结果显示:胡杨外植体最佳灭菌法为0.1% HgCl2处理6 min;愈伤组织最佳诱导培养基MS+0.50 mg/L 6-BA+0.10 mg/L NAA,诱导率>80%;愈伤组织适宜增殖培养基MS+0.05 mg/L 6-BA+0.05 mg/L NAA,增殖程度最好。

Abstract:Populus euphratica is a valuable forest resource in the desert of northwest China. However, it is difficult for Populus euphratica to take root in normal reproduction. In this paper, the branches of P. euphratica were considered as explants material, through selecting the best sterilization method and optimizing the medium to establish a culture system for induction and proliferation of callus of P. euphratica. The results showed that the optimum sterilization method for the branches of P. euphratica was 0.1% HgCl2 for 6 minutes. The optimum medium for callus induction of the branches of P. euphratica was MS+0.50 mg/L 6-BA +0.10 mg/L NAA, and induction rate was over 80%. The optimum medium for callus proliferation of P. euphratica was MS+0.05 mg/L 6-BA +0.05 mg/L NAA, and has the best degree of proliferation.
