小麦成熟胚高频再生基因型筛选及再生体系研究

摘    要:以目前生产上主栽或主推的、具有优良农艺性状的18个黄淮麦区冬小麦品种(系)和2个西北春麦区春小麦品种为材料,对它们成熟胚再生能力进行比较,从中筛选出再生频率高的基因型,并探讨不同接种方法、麦草畏质量浓度、细胞分裂素和干燥处理等对小麦成熟胚愈伤组织诱导和分化频率的影响。结果表明,小麦成熟胚愈伤组织诱导率普遍较高,为66.7%~100.0%,而分化率普遍较低,为3.6%~35.8%。基因型间差异显著,其中愈伤组织分化率较高的基因型有泛麦8号、周麦27和邢麦6号,绿苗分化率超过30%;采用胚十字形切割法接种小麦成熟胚愈伤组织分化率较高,达到54.5%~73.3%;诱导培养基中麦草畏的诱导效果显著优于2,4-D。在麦草畏质量浓度为2~4mg/L时有利于愈伤组织诱导和分化;成熟胚愈伤组织转入不含激素或含有2mg/L 6-BA或KT的分化培养基中时,分化率分别达到38.5%,45.8%和37.0%,较其他细胞分裂素和质量浓度处理分化率高,而愈伤组织分化前干燥处理6h和未干燥处理的差异不显著,干燥12h分化率明显降低。

Abstract:In this study,in order to screen out high frequency regeneration genotypes,20 wheat lines with excellent agronomic traits were selected to study the mature embryo regeneration abilities. Among these wheat lines,18 winter wheat lines are from Huanghuai wheat area and two spring wheat lines are from Northwest China spring wheat area. The effects of different inoculation methods,concentrations of dicamba,cytokinin and drying treatment on callus induction and differentiation of mature wheat embryo culture were discussed. The results showed: The callus percentage of wheat mature embryos is generally higher in 66.7%-100.0% ,but the differentiation rate is generally lower in 3.6%-35.8%. There were significant differences among different genotypes. The genotypes with higher green shoots percentage are Fanmai 8,Zhoumai 27 and Xingmai 6,the green shoots percentages are higher than 30%; the differentiation rate of wheat mature embryos with cross cutting is 54.5%-73.3%;the effect of adding dicamba in the induction culture medium is obviously better than 2,4-D. When the dicamba concentration is 2-4 mg/L,the induction culture medium is good for callus induction and differentiation; callus from mature embryos transferred in the medium without hormones or containing 2 mg/L of 6-BA or KT,the differentiation rate reached 38.5%,45.8% and 37.0%,respectively,which are higher than other treatment and concentration of cytokinin. It was no significant difference between 6 h desiccation and no desiccation before differentiation culture,but the differentiation rate was reduced obviously for 12 h desiccation before differentiation culture.
