硒对氧化应激小白鼠肝脏酶活的影响

摘    要:为研究硒对氧化应激小白鼠肝脏酶活的影响,选取45日龄36只健康昆明品系雄性小白鼠,按体重相近的原则随机分为3组:1组为对照组(硒的水平0.3mg·kg-1 W,腹腔注射10mg·kg-1 W的生理盐水);2组为低硒应激组(硒的水平0.3mg·kg-1 W,腹腔注射D-半乳糖150mg·kg-1 W);3组高硒应激组(硒的水平0.6mg·kg-1 W,腹腔注射D-半乳糖150mg·kg-1 W)。试验结束后,屠宰所有试验小白鼠,摘除眼球采血,分离出肝脏,测定MDA、T-AOC、GSH-PX、LDH、ALT、ASTD的活性。结果表明:与对照组相比,低硒应激组GSH-PX活性、T-AOC水平极显著降低,MDA浓度极显著升高;高硒应激组GSH-PX、T-AOC含量差异不显著。低硒应激组与高硒组之间ALT和ALP活性差异显著。与对照组相比,低硒组ALT活性显著,ALP活性极显著增高,高硒应激组LDH、ALT和ALP活性差异不显著,低硒应激组和高硒应激组之间差异显著。0.6mg·kg-1 W硒对D-半乳糖诱导的肝脏损伤有明显的保护作用,这种保护作用可能与硒提高抗氧化酶活性、增强机体的抗氧化防御能力有关。

Abstract:The experiment was conducted to study the effect of selenium on the enzyme activities in liver of the oxidativestressed mice.This study selected 36 healthy kunming strain male mice at the age of 45 days.Animals were randomly divided into three groups according to the principle of similar weight:the first group was for the control group (receiving 0.3 mg· kg^-1 · W Selenium and intraperitoneal injection of 150 mg · kg^-1W D-galactose),the second group was for the low selenium stress group (receiving 0.3 mg · kg 1W Selenium and intraperitoneal injection of 150 mg · kg^-1W D-galactose),the third group was for the high selenium stress group (receiving 0.6 mg · kg^-1 W Selenium and intraperitoneal injection of 150 mg · kg^-1W D-galactose).After the establishment of animal modle,all mice were slaughtered,and blood was collected by eyeball removal.Liver was quickly isolated for the determination of T-AOC,GSH-PX,LDH,ALT,and AST activities and MDA content.The results were as followed:compared to Group 1,the activities of GSH-PX and T-AOC decreased(P〈0.01),and MDA content increased in Group 2 (P〈0.01) ; in Group 3,compared to Group 1,no difference occur in GSH-PX and T-AOC activities and MDA content; the comparison of Group 2 to group 3 showed that activitive of GSH-PX and T-AOC decreased(P 〈0.05),and MDA content increased(P〈0.05).Compared with Group 1,compared to,the activities of LDH,ALT decreased(P〈0.01),and ALP activity increased in Group 2(P〈0.01),while in Group 3 activities of LDH,ALT and ALP presented no difference.Compared to Group 3,activities of LDH,ALT and ALP showed no change in Group 2(P〈0.05).In conclusion,the protective effect of selenium on D-galactose induced liver injury was obvious,and this effect may be related to enhancement of the body antioxidant ability by selenium supplement.
