摘    要:针对根结线虫16D10基因和花生△12脂肪酸脱氢酶(FAD2)基因的核酸序列设计了两条含有GUS内含子的RNA干扰结构序列.以pBS-T质粒为中间载体,将这段序列连接进植物转化载体pRI 101-AN中,获得针对两个基因的RNAi载体.经SalI和KpnI双酶切验证,目的序列已准确插入目标载体.此载体可用以培育抗根结线...

Abstract:Two RNAi sequences with a GUS intron were designed and synthesized on the basis of nucleotide sequences of root-knot nematode(RKN) 16D10 gene and Arachis hypogaea Δ12 fatty acid desaturase(FAD2) gene.The RNAi sequences were ligated into pRI 101-AN plasmid,a plant transformation vector,via pBS-T vector.Target sequence was confirmed to be inserted into the vector correctly by double digestion with SalⅠand KpnⅠ.The RNAi constructs can be used to breed high oleate/RKN resistant peanut varieties.
