南瓜白粉病不同病情等级下叶际细菌群落结构和多样性

摘    要:南瓜白粉病是一种危害严重的真菌病害,研究不同病情等级下细菌群落组成的异同对进一步研究南瓜白粉病的生物防治具有重要意义。采集来自3个不同发病等级的南瓜白粉病叶片(R1、R2和R3),运用高通量测序方法分析比较了不同发病等级的南瓜叶片的细菌群落结构和多样性,发现三个不同病情等级下细菌群落结构存在显著差异。在门水平上,三个病情等级下优势门均为Actinobacteria(放线菌门)、Bacteroidetes(拟杆菌门)、Proteobacteria(变形菌门)和Firmicutes(厚壁菌门);在属水平上,R1病情等级下优势属是Methylobacterium、Sphingomonas和Acinetobacter,R2病情等级下为Acinetobacter、Kluyvera和Pantoea,R3病情等级下为Methylobacterium、Kluyvera和Pantoea。南瓜白粉病菌可以改变南瓜叶际细菌群落结构,影响细菌群落多样性。Kluyvera、Pantoea和Erwinia的相对丰度与病情等级呈正相关,随着病情严重发生,丰度得到增强,表明它们可能与白粉菌存在协同作用。这些结果对进一步研究南瓜叶际微生态从而防控白粉病提供了科学参考依据。

Abstract:Pumpkin powdery mildew is a serious fungal disease. Investigation of bacterial community and composition on pumpk in leaves with different severities of powdery mildew will certainly help improve its biological control.The pumpkin leaves with 3 different severities (R1, R2 and R3)of powdery mildew were collected and the bacterial community structures and diversities were analyzed by high-throughput DNA sequencing.Results showed significant differences of bacterial community structures among 3 different severity levels. At the phylum level, the dominants were Actinobacteria, Bacteroidetes, Proteobacteria and Firmicutes under all 3 severity levels.At the genus level,the dominants in R1 were Methylobacterium, Sphingomonas, and Acinetobacter;the do-minants in R2 were Acinetobacter, Kluyvera and Pantoea; and the dominants in R3 were Methylobacterium, Kluyvera and Pantoea.The bacterial community structures and diversities on pumpkin leaves could be affected by powdery mildew. The relative abundances of Kluyvera, Pantoea and Erwinia were positively correlated with di-sease severities,indicating a synergistic effect of these bacteria and powdery mildew pathogen. The above results provide a theoretical basis for further study of the microecological management of powdery mildew on pumpkin.
