水仙潜隐病毒病病原鉴定

摘    要: 在中国水仙主产区水仙病毒病的系统调查中,从无症和褪绿斑驳的病叶中分离到3个分离物NC、Ⅱ-3和Ⅹ-3.经人工根接和汁液摩擦接种在昆诺藜和千日红上,表现为局部枯斑;在番杏和苋色藜上,表现为局部褪绿或局部褪绿白斑;在克利夫兰烟和菜豆上产生系统褪绿;而对曼陀罗、豇豆和裂叶牵牛则不侵染。电镜观察病原病毒为635~660nm×13nm的线状粒体。体外抗性测定表明:TIP为65~70℃、DEP为10-2~10-3,而LIV为3天。ELISA测定结果与来自荷兰水仙上的NLV标准抗血清起阳性反应。这些结果表明,3个分离物为同一病原,均属水仙潜隐病毒(NLV)。

Abstract:An investigation of virus diseases of narcissus(Narcissus tazetta var. Chinensis) was made in Fujian during 1985~1988.Three isolates,NC,Ⅱ-3 and Ⅹ-3,were obtained from narcissus plants with mottled and symptomless leaves.The three isolates caused the same symptoms on diagnostic hosts and all showed flexuous filamentous particles C. 635~660 nm x 13nm. The virus in crude sap from phaseolus vulgaris was inactivated at 65~70℃ for 10 min. and had a dilutionend point between 1:100 and 1:1000 and longevity in vitro of 3 days at room temperature. Crude sap from P. vulgaris and narcissus leaves reacted positively to the antiserum of NLV(from Netherlands) by ELISA. The three isolates were, therefore, identified to be the same virus i. e. narcissus latent virus.
