| 摘 要： | 目的]研究葛根素在大鼠体内的药代动力学规律。方法]大鼠灌胃给予葛根提取物4.285 g/kg（相当于葛根素300 mg/kg）后,采用HPLC测定不同时间点大鼠血浆中葛根素的含量,绘制各时间点平均血药浓度-时间曲线,采用3P97药动学软件对数据进行处理。结果]葛根素在0.11~17.80μg/ml线性关系良好（r=0.999 9）,其在大鼠体内的药代动力学行为符合二室开放模型,主要药动学参数：t1/2（ka）为（9.719±2.765）min,t1/2α为（16.038±4.211）min,t1/2β为（123.222±20.886）min,T（peak）为（30.679±8.305）min,C（max）为（1.178±0.575）μg/ml,AUC为（218.816±81.526）μg.min/ml,CL/F（s）为（1.515±0.492）L/（kg.min）,V/F（c）为（198.197±66.301）L/kg,Ka为（0.076±0.020）L/min,Lag time为（0.655±0.732）min。结论]该测定方法专属、准确、灵敏,适用于葛根素的药动学研究,为合理利用药用植物葛根提供了参考。

| Abstract: | Objective] The aim was to study the pharmacokinetics rule of puerarin from pueraria in rats.Method] The puerarin content in rats blood at different time points was determined after 4.285 g/kg pueraria extracts were used for the lavage of rats(equivalent to 300 mg/kg puerarin),to draw drug-time curves at different time points,in addition,the datas were calculated based on 3P97 pharmacokinetics Software.Results] A good linear relationship of puerarin was obtained in the range of 0.11 and 17.80 μg/ml(r=0.999 9),and the pharmacokinetics of puerarin in rats were fitted to two compartment open model,the main pharmacokinetic parameters were as flollws: t1/2(ka)=(9.719±2.765)min,t1/2α=(16.038±4.211)min,t1/2β=(123.222±20.886)min,T(peak)=(30.679±8.305)min,C(max)=(1.178±0.575)μg/ml,AUC=(218.816±81.526)μg·min/ml,CL/F(s)=(1.515±0.492)L/(kg·min),V/F(c)=(198.197±66.301)L/kg,Ka=(0.076±0.020)L/min,Lag time=(0.655±0.732)min.Conclusion] The HPLC method was selective,accurate and sensitive.Thus,it is suitable for pharmacokinetic studies,which will provide references for the reasonable use of pueraria. |
