利用SSR标记鉴定玉米杂交种南校18号种子纯度

摘    要:以南校18号玉米杂交种及其亲本自交系为材料,对胚芽和干种子的DNA进行提取,对24对SSR引物进行筛选,获得适合该杂交种纯度鉴定的SSR引物。结果表明,在快速检测中,从干种子胚提取DNA更加便捷、快速,无DNA降解,可用于PCR扩增反应;24对SSR引物中,有3对特异性引物6mmc0241、umc2025、phi323152可用于南校18号杂交种和亲本自交系的纯度鉴定。因此,应用SSR标记结合单籽粒DNA快速提取方法,可以快捷、准确地鉴定玉米杂交种种子纯度。

Abstract:In this study,hybrid maize Nanxiao 18 and its inbred line parents were used as materials,and DNA from embryo and embryonic bud of seed were extracted and detected by 24 pairs of primers of SSR to obtain suitable primers for identifying seed purity of the hybrid.The results showed that extracting DNA from embryonic bud of dry seed with rapid detection method was convenient and rapid,and the extracted DNA had no degradation and could be used for amplifica-tion reaction of PCR.Moreover,among 24 pairs of primer...
