洪山鸡和麻城绿壳蛋鸡禽内源性反转录病毒基因检测与分析

摘    要:为了解禽内源性反转录病毒(AERs)在湖北地方鸡种洪山鸡和麻城绿壳蛋鸡基因组中的存在状况,利用禽内源性反转录病毒EAV、ev loci和ev/J特异性引物对两种鸡的基因组进行PCR检测。结果表明,洪山鸡和麻城绿壳蛋鸡中均检测出EAV、ev loci和ev/J三种禽内源性反转录病毒基因序列;核酸序列同源性分析显示,洪山鸡和麻城绿壳蛋鸡基因组中的EAV、ev loci和ev/J之间的同源性分别为98.8%,99.6%和99.6%;进化树分析显示,两种鸡内的三种禽内源性反转录病毒均分别位于进化树的同一分支,表明洪山鸡和麻城绿壳蛋鸡内源性反转录病毒具有共同的祖先并且进化路径相似。洪山鸡和麻城绿壳蛋鸡的ev/J与ALV-J原型株HPRS-103同源性高于98%,且进化树处于同一分支。而与ALV-J国内分离株SD9901、YZ9901毒株的同源性较低,并且进化树位于不同分支,表明这两种鸡的ev/J可能与ALVJ原型株HPRS-103亲缘关系更近,而与国内的外源性ALV-J分离毒株SD9901、YZ9901亲缘关系较远。

Detection and Analysis of Avian Endogenous Retrovirus Genes in Hongshan Chicken and Macheng Green Shell Chicken
Abstract:In order to investigate the status of the Avian en dogenous retrovirus (AERs) in the genome of Hubei indigenous chickens, Hongshan chicken and Macheng green shell chicken, specific primers were set to detect the genomes of these two chickens. The results showed that the EAV,ev loci and ev/J were all detected in Hongshan chicken and Macheng green shell chicken. The EAV,ev loci and ev/J of Hongshan chicken and Macheng green shell chicken showed 98.8%,99.6% and 99.6%identity to each other. Phylogenetically,the EAV,ev loci and ev/J of Hongshan chicken and Macheng green shell chicken were in the same branch,which indicated that the AERs of Hongshan chicken and Macheng green shell chicken had a common an-cestor and they were in a similar evolutionary pathway. The ev/J of Hongshan chicken and Macheng green shell chicken showed more than 98% identity to ALV-J prototype strain HPRS-103 and they were in the the same branch of the phylogenetic tree. However, Hongshan chicken and Macheng green shell chicken showed lower identity to ALV-J strains SD9901 and YZ9901 which were separated in China comparing with the HPRS-103 strain. The results indicated that the ev/J of these two chickens are more closely related to ALV-J prototype strain HPRS-103 than the the exogenous ALV-J strains SD9901 and YZ9901.
