黄芩悬浮细胞培养过程中黄芩苷含量与POD活性的相关性分析

摘    要:以黄芩悬浮细胞为材料,研究其生长过程中过氧化物酶(POD)活性变化规律及其与黄芩苷次生合成之间的关系。结果表明,在黄芩悬浮细胞生长周期(21 d)内,POD活性与黄芩苷的积累呈正相关。在黄芩悬浮细胞生长周期的前6 d,悬浮细胞处于适应期,POD低水平表达,黄芩苷合成缓慢;6~15 d,悬浮细胞处于增长期,POD高水平表达,并到达最大值,期间黄芩苷迅速合成;15~21 d,悬浮细胞进入饱和期,此时POD活性降低,黄芩苷合成几乎停止。采用L9(34)正交表优化培养基中6-BA、碳源、氮源、p H的配比,筛选POD活性表达最优的培养条件为:6-BA 3 mg/L、碳源15 g/L蔗糖+15 g/L葡萄糖、氮源(NH4+)30 mmol/L、p H 5.6。

Analyses of the Correlation between Baicalin Content and Peroxidase Activity in the Scutellaria Suspension Cells
Abstract:The Scutellaria suspension cells were taken as the test material to study the relationship between the variation of peroxidase activity and baicalin secondary synthesis in the growth process of the Scutellaria suspension cells. The results showed that, during the growth cycle (21 days) of the callus of the Scutellaria suspension cells, the activity of the level of peroxidase and the accumulation of baicalin was positively correlated. During the the former 6 days of the growth cycle of the Scutellaria suspension cells, the suspension cells was in its adaptive phase. The activity of the peroxidase expressed at a low level. The synthesis of baicalin was slow. During 6 to 15 days, the suspension cells were in its growth phase. The activity of peroxidase express at high levels and reaches its maximum, at the same time baicalin rapidly synthetised. During 15 to 21 days, the suspension cells was in its condition period, while the activity of peroxidase reduced. The synthesis of baicalin al-most stopped. L9(34) orthogonal table was used to optimize the ratio of medium 6-BA,carbon sources,nitrogen sources, and the pH. Here is the optimal culture selected for the expression of peroxidase activity:6-BA 3 mg/L;carbon source 15 g/L su-crose +15 g/L glucose; nitrogen source (NH4+) 30 mmol/L; pH 5.6.
