家兔RAPD分析反应体系的优化
摘    要:以家兔的基因组DNA 为模板,研究影响家兔RAPD 扩增的因素。实验结果表明,Taq酶、随机引物和Mg2 + 浓度及模板DNA 的纯度对RAPD 扩增影响较大。本实验建立了适于家兔RAPD 分析的反应体系:反应体积为20μl,其中含有1 ×扩增缓冲液,2 .0m mol/L MgCl2 ,0 .1m mol/LdNTP,0-2μmol/L 随机引物,1 .5UTaq 酶,90ng 模板DNA

Optimization of RAPD Reaction System for Rabbit Genetic Analysis
Abstract:The study on factors affecting amplification of RAPD for rabbit was carried out by using rabbit genomic DNA as template. The result showed that the concentrations of Taq polymerase, random primer, Mg 2+ and the purity of template DNA played an important role in RAPD analysis.A technical system for RAPD analysis in rabbit was established: in 20μl reaction solution, containing 1×buffer, 2 0mmol/L MgCl 2, 0.2μmol/L primer, 0.1mmol/L dNTP, 90ng template DNA, 1 5UTaq polymerase.
