枸杞叶枯病拮抗芽孢杆菌的筛选、鉴定及防效

摘    要:为获得对枸杞叶枯病有较强拮抗作用的芽孢杆菌,从农田土壤中,分离、纯化得到151株芽孢杆菌,经病原真菌定向筛选得到1株对枸杞叶枯病菌拮抗活性较好的菌株LKYLW-1,其对枸杞叶枯病菌的抑菌带宽15.2 mm,采用平板对峙法、孢子萌发法和田间药效试验测定LKYLW-1的抑菌作用。结果表明:菌株LKYLW-1对枸杞叶枯病菌菌丝有致畸作用;菌株LKYLW-1发酵液对孢子萌发有抑制作用,EC50为9.35 mL·L-1;该菌株对枸杞叶枯病具有较好的治疗和预防效果。通过形态学特征、理化分析及16S rDNA序列分析对菌株LKYLW-1进行了鉴定,检测了其代谢产物,研究发现菌株LKYLW-1为萎缩芽孢杆菌Bacillus atrophaeus,其在代谢过程中可产生纤维素酶、蛋白酶和淀粉酶等抑菌物质;证明菌株LKYLW-1在枸杞叶枯病防治中具有潜在的利用价值。

Abstract:In order to obtain bacillus which has strong antagonistic effect on leaf blight of Lycium barbarum,151 strains of bacillus were isolated and purified from farmland soil,and found a strain LKYLW-1 that antibacterial bandwidth was 15.2 mm with good antagonistic activity against leaf blight of L.barbarum.The antibacterial effect of LKYLW-1 was measured by pair culture,spore germination and the field efficacy trials.The results showed that the strain LKYLW-1 had teratogenic effects on the mycelium of the leaf blight of L.barbarum.The fermentation broth of strain LKYLW-1 could inhibit the spore germination,and the EC50 was 9.35 mL·L-1.In the field efficacy trials,the antagonistic strain LKYLW-1 showed good preventive and therapeutic effects on leaf blight of L.barbarum.The strain LKYLW-1 was identified as Bacillus atrophaeus by morphological characteristics,physical and chemical analysis and 16 S rDNA sequence analysis,which could produce cellulase,protease and amylase during metabolism.So the strain LKYLW-1 has potential utilization value on the control of leaf blight of L.barbarum.
