菜豆中阿维菌素残留量的高效液相色谱检测方法及消解动态研究

摘    要:建立了反相高效液相色谱法测定菜豆中阿维菌素残留量的方法,以乙酸乙酯为提取剂,液液萃取净化,经N-甲基咪唑和三氟醋酸酐衍生化,采用高效液相色谱法-荧光检测器(激发波长365 nm,发射波长为470 nm)进行测定。同时,为确定1%阿维菌素水乳剂在菜豆中的施用安全性,通过两年两地田间试验和样品室内检测试验,研究了其在菜豆上的消解动态及最终残留量。试验结果表明:在0.05~5.0 μg/mL浓度范围内,阿维菌素的浓度与峰面积线性关系良好,线性相关系数γ为0.9997。添加浓度在0.0044~0.44 mg/kg 时,方法的平均回收率为94.35~99.42%,相对标准偏差为4.81~5.84%;以1%阿维菌素水乳剂0.45 g/m2喷药,阿维菌素在菜豆上的消解半衰期为1.4~1.7 d;施药7 d后采集的菜豆样品中阿维菌素含量均低于0.05 mg/kg (中华人民共和国农业行业标准NY1500.1.3-2007规定阿维菌素在豇豆中的最大残留限量,即MRL值,本文以此作为菜豆中阿维菌素的推荐MRL值)。本方法简便、快速、准确,适用于阿维菌素在菜豆上的残留量测定。

Determination of residue and dynamic of abamectin in kidney bean by HPLC

Abstract:A facile and sensitive method utlizing high performance liquid chromatography with fluorescence detector (HPLC-FLD) has developed for analysis of abamectin in common bean. samples were extracted with acetate, followed by derivatization with trifluoroacetic anhydride and N-methylimidazole, before analysis by HPLC-FLD of the fluoracetic detector. Fluorescence detector was employed for detection with the excition wavelength of 365 nm and emission wavelength of 470 nm. The law of residual degradation dynamic and its final residues of 1% avermectins in common bean and soil were studied by a two-year and two-field trial in Guiyang and Beijing. The half life of abamectin is 1.4~1.7 d. If the spiked level form 0.0044 to 0.44 mg/kg, the recoveries were 94.35~99.42%, and the relative standard deviations(RSD) were 4.81%~5.84%. Under conditions of 0.450 g/m2 application, the residue in commom beans was lower than recommended 0.05 mg/kg (The agricultural industry standard of the People"s Republic of China NY1500.1.3-2007 determines the maximum residue limit of avermectin in the cowpea, MRL, which is used the recommended MRL value in common bean). The method is simple and rapid with high sensitivity making it suitable for determination of abamectin in commom bean.
