薰衣草离体培养技术研究

摘    要:以薰衣草(Lavandula angustifolia)带芽茎段或顶芽为外植体,探讨不同激素种类与水平等对其愈伤诱导、不定芽增殖与不定根形成的影响,并筛选了薰衣草试管苗过渡移栽的适宜基质。结果表明薰衣草离体培养适宜的培养基分别为,不定芽启动培养基,MS+BA 1.0~2.0 mg/L+蔗糖30 g/L+琼脂6.5 g/L;继代增殖培养基,MS+BA 1.0~2.0 mg/L+IBA 0.25 mg/L+蔗糖30 g/L+琼脂6.5 g/L;生根培养基,White+IBA0.4 mg/L+蔗糖20 g/L+琼脂6.5 g/L或White+NAA 0.8 mg/L+蔗糖20 g/L+琼脂6.5 g/L。以等体积的泥炭与珍珠岩混合后作为基质驯苗,幼苗成活率可达96.9%。

Abstract:Using stem or tip bud of Lavandula angustifolia as the explant,the effects of external hormone in medium on inducement of callus,multiplication of adventitious buds,and formation of adventitious roots were studied;And suitable ground substance for transplantation of L.angustifolia seedling was screened out.The results showed that medium for callus inducement should better be MS+BA 1.0~2.0 mg / L+ sucrose 30g / L+ agar 6.5 g / L.MS+BA 1.0~2.0 mg / L+IBA 0.25 mg / L+sucrose 30 g / L+ agar 6.5 g / L was good for multiplication of adventitious buds.And White+IBA 0.4 mg / L+ sucrose 20 g / L+ agar 6.5 g / L or White+ NAA 0.8 mg / L+ sucrose 20 g / L+ agar 6.5 g / L was suitable for adventitious roots formation.Using mixed peat and vermiculite with equal volume as transplantation ground substance was contributory to the growth of seedling as the survival rate could reach 96.9%
