香石竹尖孢镰刀菌PCR检测

摘    要:采用多重引物聚合酶链式反应(Mulljplex—PCR)技术检测香石竹尖孢镰刀菌纯培养和香石竹样品,并用分离培养技术加以验证。结果表明,PCR能特异地检测出所有16个香石竹尖孢镰刀菌菌株,并证实了昆明地区的香石竹尖孢镰刀菌为2号生理小种。PCR与分离培养技术检测结果基本一致,但总体上PCR检测阳性率稍高于分离培养检测的发病率。PC能检测出接种7d后香石竹病原菌,而观察病症需20d。该项技术具有更高的灵敏度,适用于香石竹种苗的检测和病害流行学研究。

Abstract:Fusarium oxysporum f.sp. diathi, which causes carnation wilt, was detected by multiplex PCR. PCR technique succeeded in detecting 16 strains isolated in Kunming., and the strains were identified as physiological race 2 of the pathogen. The PCR procedure gave similar results to those of biological method but was more sensitive in general. The pathogenic fungi could be detected by PCR 7 days after inoculation, as compared to 20 days by symptom observation. It is thus recommended that PCR technique be employed in carnation plant testing and in epidemiological research.
