荷兰铁组织培养研究

摘    要:对荷兰铁组织培养方法进行研究,结果表明:荷兰铁顶芽经消毒处理后接种到MS+6-BA 2.0 mg/L+NAA 0.1 mg/L培养基中,可诱导出愈伤组织和不定芽;在1/2MS+6-BA 1.0 mg/L+NAA 0.1 mg/L培养基中继代培养,扩增倍数(MR值)约2.2倍;将继代培养芽丛中高3cm以上的不定芽接种到生根培养基1/2MS+IAA 0.3 mg/L中,30 d左右生根率达98.2%;炼苗20~30 d后移栽,成活率达95%以上。

Study on Tissue Culture of Yucca elephantipes

Abstract:The tissue culture of Yucca elephantipes was studied.The results showed that the terminal bud of Yucca elephantipes was planted in the culture medium MS+6-BA 2.0 mg/L+NNA 0.1 mg/L after being disinfected,then the callus and adventitious bud were induced.The expansion rate was 2.2 after being subcultured in 1/2MS+6-BA 1.0 mg/L+NAA 0.1 mg/L.Then the subculture buds long than 3 cm were inoculated in rooting medium 1/2MS+IAA 0.3 mg/L,and the rooting rate reached 98.2% after 30 days.The survival rate of transplant was high than 95% after 20~30 days adaptation.
