摘    要:Caco-2细胞源自人结肠癌细胞,体外培养时能自发地进行类似肠道细胞的形态学和生化学上的分化,获得许多小肠吸收细胞的特性,如形成微绒毛结构;在细胞表面形成良好的刷状缘;在细胞间形成紧密连接;分泌水解酶以及合成转运糖、氨基酸和药物等的载体转运系统。由培养在微孔滤膜上的Caco-2细胞构建的模型为研究营养素在小肠的吸收机理提供了一个有效且易于操作的实验手段。本文主要综述了Caco-2细胞模型的建立、特征、检测及其在氨基酸、维生素、核苷和微量元素等营养素小肠吸收机理研究中的应用。

Abstract:The Caco-2 cell line is derived from a human colorectal carcinoma.The Caco-2 cells undergo spontaneous morphological and biochemical enterocytic differentiation in culture in vitro and develop a well-defined brush border on the apical surface as well as tight cellular junctions.They also acquire many features of absorptive intestinal cells during culture,such as microvillous structures,hydrolysis enzymes and carrier mediated transport systems for sugar,amino acids,vitamins,and drugs.The well-formed Caco-2 cell monolayer model is firstly applied in the field of pharmacology.It can be used to identify drugs with potential absorption problems and also to select drugs with optimal passive absorption characteristics from series of pharmacologically active molecules generated in drug discovery programs.It is well known that the process of permeability,uptake,and transport of nutrients in intestine is one of the most important steps to investigate the absorption mechanisms of nutrients as well as their bioavailability.The Caco-2 cells cultured on micro-porous membranes in inserts contained in transwell or snapwell biochambers also constitutes an effective in vitro model for elucidating the mechanisms of nutrient absorption in intestine by numerous investigators.It offers a number of advantages including rapid assessment of absorption and metabolism of molecules;the use of cells as oppose to tissues or animals;great viability;and better reproducibility.This paper reviewed the construction,characterization,and integrity detection of Caco-2 cell monolayer model as well as its application in elucidating the mechanisms of absorption of nutrients,such as amino acids,vitamins,nucleoside and trace elements and so on in intestine.
